Tirzepatide is a once-weekly dual GIP/GLP-1 receptor agonist marketed as Mounjaro and Zepbound. The SURPASS program established efficacy in type 2 diabetes (HbA1c reductions of ~2.0-2.4 percentage points), and the SURMOUNT program established the largest weight-loss effect sizes seen in any RCT to date (up to 20.9% mean body-weight reduction at 72 weeks).
