Dulaglutide is a GLP-1 analog fused to a human IgG4 Fc fragment, dramatically extending half-life to ~5 days and enabling weekly subcutaneous dosing. The Fc-fusion construction also reduces immunogenicity. FDA-approved for type 2 diabetes glycemic control plus cardiovascular event reduction (REWIND trial, Lancet 2019). Known for simplest auto-injector device in class.

Trulicity's Fc-fusion structure provides excellent diabetes glycemic control but produces less appetite suppression and weight loss than semaglutide or tirzepatide. Dulaglutide molecular size may limit central nervous system penetration affecting appetite regulation. Trulicity is not FDA-approved for obesity; weight loss is secondary effect in diabetes treatment.
Trulicity is FDA-approved only for type 2 diabetes. Off-label prescription for weight loss alone is uncommon since Wegovy and Zepbound are specifically FDA-approved for obesity with higher efficacy. Trulicity may produce modest weight loss as a secondary benefit during diabetes treatment.
